Retatrutide is a triple agonist targeting GLP-1, GIP, AND glucagon receptors, making it the most mechanistically complex metabolic peptide currently in research. The addition of glucagon agonism introduces direct thermogenic effects (increases energy expenditure) and enhanced lipolysis on top of the appetite suppression and insulin regulation of dual agonists. Phase 2 trials showed up to 24.2% weight loss at 48 weeks, exceeding tirzepatide in preliminary comparisons.
